Intra-assay Precision (Precision within an assay) Three samples of known concentration were tested twenty-two times on one plate to assess intra-assay precision.

Inter-assay Precision (Precision between assays) Three samples of known concentration were tested six times on one plate to assess intra-assay precision.

Spike Recovery

The spike recovery was evaluated by spiking 3 levels of rat TIMP1 into health rat serum sample. The un-spiked serum was used as blank in this experiment.
The recovery ranged from 80% to 103% with an overall mean recovery of 97%.

Sample Values

Sample Matrix Sample Evaluated Range (ng/ml) Detectable (%) Mean of Detectable (ng/ml)

Serum/Plasma– Thirty samples from apparently healthy rats were evaluated for the presence of rat TIMP1 in this assay. No medical histories were available for the donors. n.d. = non-detectable. Samples measured below the sensitivity are considered to be non-detectable.

Background: TIMP1

TIMP-1 (tissue inhibitor of metalloproteinase-1) is a secreted glycoprotein that blocks metalloproteinase (MMP) activity. It circulates in complex with various MMPs to maintain them in an inactive state. It is produced by fibroblasts, osteoblasts, endothelial cells, granulosa cells, dendritic cells, vascular smooth muscle cells, adipocytes, and monocytes. TIMP-1 also protects against apoptosis in B cells, stimulates erythropoiesis and BFU-E expansion, both stimulates and inhibits tumor growth, inhibits angiogenesis, and stimulates gonadal steroidogenesis in conjuction with Cathepsin L.
