Intra-assay Precision (Precision within an assay) Three samples of known concentration were tested twenty-two times on one plate to assess intra-assay precision. Inter-assay Precision (Precision between assays) Three samples of known concentration were tested six times on one plate to assess intra-assay precision.

Spike Recovery

The spike recovery was evaluated by spiking 3 levels of human MCP-2/CCL8 into health human serum sample. The un-spiked serum was used as blank in this experiment.
The recovery ranged from 79% to 117% with an overall mean recovery of 93%.

Serum/Plasma – Thirty samples from apparently healthy volunteers were evaluated for the presence of human MCP-2/CCL8 in this assay. No medical histories were available for the donors. n.d. = non-detectable. Samples measured below the sensitivity are considered to be non-detectable.

Background: MCP-2/CCL8

MCP2 and MCP3 are two recently identified monocyte chemotactic proteins produced by human MG63 osteosarcoma cells. Both MCP2 and MCP3 are members of the C-C family of chemokines and share 62% and 71% amino acid sequence identity, respectively, with MCP1. MCP3 also shares 58% amino acid identity with MCP2. Similarly to other C-C chemokines, all three MCP proteins are monocyte chemoattractants. In addition, the three MCPs can chemoattract activated NK cells as well as CD4+ and CD8+ T lymphocytes. All three cytokines have also been shown to attract eosinophils and induce histamine secretion from basophils.
